Effects Of Antıepıleptıcs (Lamotrıgıne And Oxcarbazepıne) On Mat-Lylu Rat Prostate Cancer Cell Lıne: Prolıferatıon And Mıgratıon

Prostate cancer, which has rapidly rate of increase by advancing age among men, is a very common type of cancer. A lot of studies display that, voltage gated sodium channels (VGSC) have very important roles at prostate cancer’s development and progression. Also it was showed that, VGSC expression plays a role as an accelerator factor and some of the studies point out that VGSCs may have a potential for the diagnosis and treatment of this disease as a therapeutic marker. Evidences from cancer studies are very important for enhancing new therapy methods, establishing VGSC specific agents and for explaining their efficacy profiles.

Epilepsy is a chronic disease that is characterized by repetitive seizures and needed to be cured particularly. The antiepileptic drugs used for the treatment have different efficacy mechanisms. The new antiepileptics, lamotrigine (LTG) and oxcarbazepine (OXC) we used for the study, provide the suppresion of epileptic seizures and show their effects via Na+ channel blockade.

The aim of this study is to determine the effects of LTG and OXC onto Dunning rat prostate tumour model, highly metastatic Mat-LyLu cells’ kinetic parameters like toxcicity, proliferation and labelling index. Also it was aimed to show the effects of these agents on lateral movement of cells by comparing with the other VGSC specific blocker tetrodotoxin (TTX).

For this purpose 75 µM LTG, 0,1 µM OXC, 600 nM and 1 µM TTX were applied onto Mat-LyLu cell line for 24 and 48 hours, furthermore lateral movement of the cells were studied together with cell kinetics. According to achieved results, 75 µM LTG and 0,1 µM OXC did not produce a statistical effect (p>0,05) on proliferation and migration of cells, however they stimulated DNA synthesis. Also it was determined that, LTG and OXC did not show an obvious inhibition such as high specific blocker TTX.
