Determınatıon Of Gen Expressıon In Canola (Brassica Napus L.) Under Hypobarıc Pressure
In this study, it is aimed to perform molecular analysis of the changes as a result of different low pressure levels in canola plant (Brassica napus L.) such as Californium, Jura, Orkan and Elvis varieties.
For that reason, Canola varieties were planted and those plants grown in soil were placed in specially designed low pressure cabinets and were exposed to low pressure (40 torr) for varying periods (1, 2 and 3 days) .Total RNA isolation is done from the plants and to analyze the respond of canola varieties to low pressure levels at the molecular level, expression profiles of gene regions, OsNCED, OSABA8, OsZEP and TMAC2 were examined by RT-PCR technique. In addition, as a control of the study, β-actin gene region was used as housekeeping gene to confirm whether PZR technique is working accurate or not.
As a result of the study, when the varieties of canola plant, Californium, Jura, Okran and Elvis were exposure to low pressure, it was observed that expression profiles of the genes, OsABA8 and OsNCED changed. Because any other studies related to low pressure have not been done in our country, this study is the first and fundamental to lighten further studies.
