RESEARCH EXPRIENCE

1. Pre-doctoral

The work of my Ph.D. thesis deals with the isolation, purification and physico-chemical study on pigeon pea lectin. The lectin was purified by salt precipitation followed by affinity chromatography. The purity and molecular weight of the lectin was determined by gel filtration and SDS-PAGE. The UV-CD spectra of lectin suggest that the predominant secondary structure is β- pleated sheet. The saccharide binding specificity was investigated by hapten-inhibition of hemagglutination or glycoconjugate precipitation. The presence of various ionizable groups was determined by hydrogen-ion titration. Numbers of phenoxyl groups present in lectin were also determined by spectrophotometric method. The effect of metal ions on carbohydrate binding activity of lectin was investigated in detail.



2. Post-doctoral

This involves the scale up of biological processes for the production of recombinant proteins from E.coli, process optimization, downstream operation for protein purification. Also, optimization of solubilization process for recombinant proteins was made in order to refold them to their native structure.



3. PRESENT WORK

Currently involved in the following aspects of Structural Biology:



  1. Protein stability, Structure and Function

  2. Amyloid induction and inhibition

  3. Characterization of folding intermediates of proteins at low pH.

  4. Interaction of lectin with rhizobia.

  5. Purification and physico-chemical characterization of lectin from different sources.

  6. Protein aggregation and misfolding
