Gene Transfer Into Maize (Zea mays L.) Via Particle Bombardment Method
The main objective of this study was to optimize the conditions for the tissue culture and gene transfer into Turkish local maize (Zea mays L.) cultivars via particle bombardment.

Optimization of the tissue culture conditions is necessary for the successful plant transformation. It is known that (commonly accepted that) tissue culturing and regenerating fertile plantlets are difficult in maize since it is a monocot plant. Murashige and Skoog (MS) and Chu (N6) tissue culture mediums were used in this study. All mediums were arranged for the optimum plant growth (and plant morphogenesis).

Several maize cultivars were tested to determine the best local maize lines for the alternate crop in the field, and ADK 310, ADK 451, ADK 455, ADK 599, ADK 694 and ADK 737 lines were selected. These lines were developed by Sakarya Agricultural Research Institute (STAE). ADK 737 was determined the most suitable line for transformation studies.

Gene transfer via particle bombardment is one of the most efficient methods for maize transformation. In order to optimize particle bombardment method, some parameters were tested such as once and twice shooting repetition, 6 cm and 9 cm shooting distance, 650 psi and 1100 psi pressure for helium gas, and 1, 4 and 20 hours osmotic treatment. Optimum parameters for maize transformation via particle bombardment were described.

Gene cassettes used in this study were provided from the TUBITAK , grant number TOVAG-109O664 that is directed by Assoc. Prof. Dr. Selma ONARICI. Plasmids with bar selective gene were used. Transformant plants produced from transformation and regeneration were detected with PCR analysis.
