and ChromosomeDamage In Human Perıpheral Blood Lymphocytes
Parkinson disease (PD) is a degenerative processes which mainly effects basal ganglions as substansia nigra and other peduncle pigmented neuron. During the aging processes, telomers might get shorter and also chromosomal damages caused by hereditary and environmental factors may have proportional role on aging. Micronucleus technique is one of the chromosome damage assessment methods. Micronucleus (MN) is a small nucleus structure consisted from a whole or sectional chromosomes which are damaged and doesn't join to the new nuclei during the cell division.
In these thesis project, the relationship of chromosome aberrations in human peripheral blood lymphocytes with Parkinson disease was investigated by MN formation. In addition to measuring spontaneous MN formation, MN frequencies and proliferation indexes (PI) after implementation of hydrogen peroxide stress in vitro were also determined. This enabled to study directly cytogenetic affects of oxidative damage by application of hydrogen peroxide stress independently from environmental factors and dietary habits
This project was performed with thirty women and man patients with Parkinson disease and same age matched healthy individuals (control group).Peripheral blood samples from individuals were drawn into two separate sterilized lithium-heparin tubes and one tube was left as a control for spontaneous MN frequencies and the other was applied hydrogen peroxide. Paired t-test was applied for the comparison of spontaneous and hydrogen peroxide induced micronucleus frequencies for each individual in both patients and control groups.
