Determination of Sukkula Retrotransposon Movements
In Barley (Hordeum Vulgare L.) Tissue Culture
Retrotransposons are dynamic elements of the genome and exist with high percentage in genome of many organisms. Although majority of retrotransposons are autonomous, there are also non-autonomous retrotransposons, which use other elements gene products for movements. Some stress conditions might have a stimulating effect for activation of retrotransposons. Tissue culture could be accepted as one of these stress factors. In this study, Sukkula (a non-autonomous retrotransposon) polymorphism was investigated in calli with different culturing time (40 and 80 day-old) which formed from the same embryo in MS medium supplemented with 3 mg/l dicamba. Mature barley embryos (Hordeum vulgare cv. Tokak 157/37) were cultured for callus formation and sub-cultured in every 20 days. Three experiment sets were constructed to determine the polymorphism between individual calli formed from different embryo in same culture time. Polymorphism was detected using Inter-Retrotransposon Amplified Polymorphism (IRAP) and Inter Primer Banding Side Amplification (iPBS) techniques with two different Sukkula specific primers.
Non-cultured 5 mature embryos were used as control. While up to 14% polymorphic PCR bands were obtained from non-cultured embryos, 61% and 0% polymorphic bands were observed with IRAP analyses in 40 and 80 day-old calli, respectively. iPBS analyses revealed more polymorphism ratio up to 29% in non-cultured materials, and up to 58% and 70% in 40 and 80 day old calli materials, respectively. Results showed that tissue culture conditions and age of calli affected Sukkula retrotransposon movements and all individuals did not show the same effect. As it is known, this is the first report on employment of Sukkula basedIRAP and iPBS applications in barley in terms of callus development.
