In our project we try to characterize the effect of IP6 and Ins as a potential adjuvant in carcinoma treatment and find the most suitable concentration and incubation period with maximum anti-cancer effect. There is some evidence that myo-inositol (Ins) can potent anticancer effect of IP6. We chose tree colorectal cell lines HT29, SW480, SW620 with different malignant potential. Cultivation was arranged for 24, 48 and 72 hours. We tested IP6 and Ins alone or in equimolar ratio in concentrations 0,2; 1 and 5 mM for both chemicals. The concentrations and incubation periods were chosen according to low toxicity of the tested substance that was observed in long-term clinical study. We measured activity of caspase-3 as an apoptotic marker. We assessed BrdU (proliferation in S-phase), WST (metabolic activity) and Brilliant Blue (proteins content). Increase of caspase-3 activity: the highest increase was observed in concentration 5 mM IP6 or IP6/Ins in HT29 cells and in concentration 0,2 mM IP6 or IP6/Ins in SW480 cells. Decrease of proliferation: all concentrations of IP6 or IP6/Ins decrease proliferation in all cell lines. Decrease of metabolic activity: IP6 and IP6/Ins decreased metabolic activity in HT29 and SW620 cells only in the highest concentration, in SW480 cells in all concentrations except 0,2 and 1 mM IP6 or IP6/Ins in 24-hour incubation. Decrease of protein content: the results correspond to the results of WST. This study demonstrates ability of IP6 alone or in combination with Ins to reduce the proliferation rate of tested cell lines and increases the proapoptotic effect. The chosen cell lines respond differently to concentrations tested.
