The bacteria within : epigenetic study of transposable elements in endosymbiotic insects
Transposable elements (TEs) are DNA sequences capable of replicating within the genome potentially causing harmful mutations. Many TE regulatory mechanisms exists, including epigenetic pathways : histone post-translational modifications and chromatin remodelling factors, DNA methylation and small RNAs. While such mechanisms are well studied in model species as Drosophila melanogaster, Arabidopsis thaliana or even in human and mouse, very little is known in other non model species, specially in endosymbiotic insects. Symbiosis is an association between two or more organisms and it occurs at different levels of integration. The most integrated association is called endosymbiosis, in which the endosymbiont is secluded in host specialized cells, the bacteriocytes, and is transmitted maternally through generations. While it is well admitted that endosymbiosis is a driving force in species adaptation and evolution, very little is known about its impact on host genome structure, transcription, and TE regulation.
Weevils are a common crop pest causing an estimated worldwide loss of hundreds of millions of dollars and hence their study is crucial for ecologic and economic reasons. Sitophilus oryzae, the rice weevil, and Sodalis pierantonius, the endosymbiont, share a very recent relationship that is illustrated by an abundance of TEs in the bacteria and host genome. Bacteriocytes are present throughout development in ovaries where they can be passed to the next generation. Furthermore, TE copies are tightly regulated in the germline, where small interfering RNA pathways are specialized in TE control.
We hypothesize that the presence of bacteria in the ovaries is able to affect TE regulation. Hence, the master student will perform knock downs of key actors of TE epigenetic regulation and follow bacteria load, bacteria localization and TE expression in germ cells. The student will also localize the proteins necessary for TE regulation in the different ovarian tissues.
We are looking for a highly motivated master student, that will learn how to manipulate weevils, perform knock downs and quantitative PCRs along with FISH and immunostaining assays. The student will be closely working with the two co-supervisors (BF2i and LBBE) and other members of the BF2i team. We expect a proactive person who enjoys being part of a team. Perspectives will involve molecular biology methods and genome-wide analysis. BF2i laboratory hosts a diverse multinational team, studying two main model systems, weevils and aphids, and it is at the frontier of the fundamental and applied research. LBBE is a evolutionary biology lab, with multidisciplinary teams from modeling to fonctional genomics.
