Movements Of Nıkıta Retrotransposon In Barley (Hordeum Vulgare L.) Tıssue Cultures
In this thesis project, Nikita retrotransposon movements were investigated in barley (Hordeum vulgare L.) tissues culture conditions using IRAP (Inter-Retrotransposon Amplified Polymorphism) molecular marker technique. For this purpose, embryos were dissected from mature barley seeds and callus formation was encouraged in Murashige Skoog (MS) medium. Samples were obtained from 30, 60 and 90 days old calli cultures and were used for genomic DNA isolation. The isolated genomic DNAs were used as template and IRAP-PCR (Polimeraz Chain Reaction) was performed with two different Nikita specific primers (N57 ve N2647). PCR products were separated and analyzed in Polyacrylamide Gel Electrophoresis (PAGE)
Results obtained from this thesis project has shown that polymorphisms depend on Nikita retrotransposon movements in different aged calli. Besides, polymorphisms between different calli in same ages showed that Nikita movements might not be the identical in each individual and might be random.
Studies on retrotransposon movements in barley tissue culture, are limited. Results obtained from this thesis, are expected to contribute to understanding of the effects of epigenetic changes on callus formation.
