Effects of Edaravone on Liver Injury by Valproic Acid in Rats
Reactive oxygen species (ROS) play a vital role in valproic acid induced liver damage. ROS damage numerous biochemical structures particularly lipids, proteins and nucleic acids. Antioxidant defense systems and their components, including antioxidant enzymes, foods and drugs are important in preventing the toxic and disease causing effects of oxygen-derived free radicals. The purpose of the present study was to investigate the protective effects of edaravone against valproic acid-induced liver injury in rats. Male Sprague-Dawley rats were used in the study. The rats were randomly distributed into 4 groups. Group I; control rats. Group II; rats receiving intraperitoneally 0.5 g/kg valproic acid, daily for 7 days. Group III; rats receiving 30 mg/kg edaravone for 7 days, intraperitoneally, daily Group IV; rats receiving 0.5 g/kg valproic acid, 2 h prior to the administration of 30 mg/kg edaravone for 7 days, intraperitoneally, daily. At 16 h after valproic acid and edaravone administration all the animals were sacrificed under ether anestesy. The administration of valproic acid caused a decrease in levels of liver tissue glutathione content, paraoxonase, biotinase activity and an increase in the serum aspartate aminotransferase, alanin aminotransferase, alkaline phosphatase activities and bilirubine content, tissue lipid peroxidation and protein carbonyl content, myeloperoxidase, xanthine oxidase, adenosine deaminase, sodium potasium ATPase, sorbitol dehydrogenase, glutamate deyhdrogenase, DT diaphorase, arginase and tromboplastic activities. Treatment with edaravone reversed these effects. These results suggested that the protective effects of edaravone can be attributed to its reducing effect on oxidative damage.
