Abstract: Vascular bed reactivity to exogenous LTD4 and blockade of these responses by a selective LTD4 receptor antagonist, SK&F 104353, were studied. Yorkshire pigs (N = 13; 25.8 +/- 1.4 kg) were anesthetized with isoflurane, and monitored for cardiac output (CO), mean pulmonary artery pressure (MPAP), pulmonary capillary wedge pressure (PCWP), right ventricular stroke work (RVSW), mean arterial pressure (MAP), renal artery blood flow (RABF), pulmonary vasculature resistance (PVR), renal vascular resistance (RVR), and systemic vascular resistance (SVR). LTD4 (0.03, 0.1, 0.3, 1, 3 and 10,0-mu-g/kg) was given intravenously. The MPAP, MAP, and RABF were recorded at baseline and for 20 min after the injection of LTD4. SK&F 104353 was infused (3 mg/kg/h, i.v.) and the dose-response to LTD4 was repeated. LTD4 induced changes in the MPAP, PCWP, MAP, and RABF. SK&F 104353 attenuated LTD4 induced changes in the MPAP, PVR, RABF and RVSW. MPAP was increased 123 +/- 24% and 115 +/- 10%, respectively, in pigs given 3.0 and 10-mu-g/kg doses, and increased to only 36 +/- 9% at a 10-mu-g/kg dose of LTD4 in animals pretreated with SK&F 104353 (P < 0.05). MPAP was not increased at the 3.0-mu-g/kg dose, and increased to only 36 +/- 9% at a 10-mu-g/kg dose of LTD4 in animals pretreated with SK&F 104353 (P < 0.05). These changes were paralleled by a reduction in PVR. RABF was decreased 76 +/- 5% and 83 +/- 7%, respectively, at the 3.0 and 10-mu-g/kg dose of LTD4. Following pretreatment with SK&F 104353 the decrease was 33 +/- 7% and 67 +/- 5%, respectively, at the 3.0 and 10-mu- g/kg doses of LTD4. RVSW also was increased 166 +/- 64% and 178 +/- 75%, respectively, at the 3.0 and 10.0-mu-g/kg doses of LTD4. SK&F 104353 completely inhibited the change in RVSW at the 3.0-mu-g/kg dose and attenuated the increase at 10-mu-g/kg to 13 +/- 8%. CO and CVP were not significantly changed by either LTD4 or SK&F 104353 at 1, 10, or 20 min. The LTD4 induced changes in MAP, SVR, and PCWP were not blocked by SK&F 104353. In summary: 1) Porcine renal, pulmonary and systemic circulations are highly sensitive to LTD4 and 2) SK&F 104353 diminishes LTD4 induced changes in RABF, RVR, RVSW, PVR, and MPAP, but not MAP, SVR and PCWP. The results suggest that subsets of LTD4 receptors may exist in various vascular beds

Abstract: An experimental study has been conducted to evaluate and compare the use of fumigated diesel fuel or gasoline as supplementary fuels for a naturally-aspirated, four-stroke diesel engine with a swirl-combustion chamber. The supplementary diesel fuel or gasoline is introduced together with the aspirated air (fumigation) in various proportions with respect to the main diesel fuel, which is injected in the usual manner. The influence of fuel/feed ratios (supplementary or main feed), for a large range of loads, has been examined on fuel consumption, pressure diagrams, exhaust smokiness and exhaust-gas emissions (nitrogen oxides, hydrocarbons and carbon monoxide). Knocking limits have been determined. The differences in the measured performance and exhaust-emission parameters from baseline engine operation, when using either supplementary diesel fuel or gasoline fumigated in the intake air, are determined and compared. Our study shows promise for this approach and indicates that above approximately 60% of maximum load, there is high smoke reduction with only a slight change in specific fuel consumption, when using either one of the supplementary fumigated fuels. Examination of gaseous pollutant levels shows involved relations with respect to load and fuel proportions. Theoretical aspects of the supplementary fuel-mode (fumigation) of combustion are used to explain the observed engine behaviour

Abstract: In adult respiratory distress syndrome (ARDS), the pulmonary artery hypertension is followed by increases in right ventricular diastolic and systolic volumes and a decreased ejection fraction. The stroke volume is preserved by the Frank-Starling mechanism as preload increases, even in the presence of severe pulmonary artery hypertension. In contrast, if there coexists a depression of the right ventricle contractility, as during right ventricular contusion, septic shock or a viral myocarditis, the compensatory Frank-Starling mechanism, that maintains right ventricular pump function, seems limited. Thus, it appears that the contractile state of the right ventricle can influence the clinical course of ARDS. In addition, patients with ARDS require mechanical ventilation with positive end-expiratory pressure (PEEP), which has a detrimental effect on right ventricular loading conditions. Most investigators agree that the most important effect is a decreased right ventricular preload, secondary to the increased pleural pressure due to PEEP. However, in patients with severe pulmonary artery hypertension, the PEEP-induced increase in right ventricular afterload may become more preponderant, and inotropic support to maintain right ventricular stroke volume may be necessary
