Proteomes in Salt Stressed Populus tremula L. (tremulous poplar) X Populus alba L. (white poplar) 717-1B4 Proteomic Investigations
Populus (Poplar) is a woody plant used as a model species and important for studying the specific features for the trees such as wood formation, long-term multi-year growth, autumn senescence, budding, seasonal accepted, flowering control, biotic interactions and adaptation. In the related investigations, Populus tremula x Populus alba (717-1B4) genotype was preferred because it can be transformed by Agrobacterium tumefaciens.
Salt stress is a major abiotic stress that limits productivity in many regions of the world. In this research, an integrated physiological and proteomic approach was used to understand the salt stress responses of Populus tremula x P. alba (717-1B4). Cuttings of a one-year-old clones were treated with 75 and 150 mM NaCl concentrations for 51 days. During the treatment, total leaf area, stem height, total number of leaves and the diameter of radial growth were measured and several differences were obtained. CO2 assimiliation rate and stomatal conductance to water vapour were significantly decreased after the NaCl treatments. The total proteins from the roots, leaves and cambial samples of NaCl treated and untreated plants were extracted and separated by Two-Dimensional Electrophoresis. At the end of the 2-D electrophoresis, 600 spots for root samples and 900 spots for leaf samples were observed per each gel. These proteins should be investigated in the future studies. At the end of the stress time courses, the elemental distribution in the roots, cuttings, stems and leaves of the young trees were measured and compared to the control many differences have been seen. For the time being, only a few studies in the literature deal with salt stress effects on poplars plants. The results obtained in this study will contribute the knowledge about abiotic stress response in woody plants.
